Our lab is interested in understanding the molecular mechanisms underlying epigenetic regulation. Genetic information in eukaryotic cells is stored in the form of chromatin, with nucleosome as repeating unit. Each nucleosome is comprised of 147-bp DNA wrapped around an octamer of histone proteins. DNA methylation and posttranslational modifications of histone proteins can significantly influence chromatin structure and function, resulting in heritable change of gene expression without changes in the genetic code, the DNA sequence. These epigenetic modifications are involved in a broad spectrum of cellular activities, including gene transcription, DNA repair, DNA replication and cell differentiation. Patterns of each epigenetic mark are dynamically regulated by a set of chromatin modifiers, whose dysregulation is linked to many human diseases, such as cancer. By using X-ray crystallography and NMR spectroscopy, combined with molecular biology and biochemistry, we aim to elucidate the molecular mechanism of key chromatin modifying enzymes, in particular their structure-function relationships and how their enzymatic activities are regulated by other epigenetic players.
